ABSTRACT

Objective:To investigate the arousal mechanism after sevoflurane anesthesia using orexinergic modulation in dorsal raphe nucleus(DRN) by optogenetic and chemogenetic techniques in rats.Methods:Forty-five healthy male Hcrt-Cre rats, aged 10-12 weeks, weighing 220-250 g, were divided into 6 groups by the random number table method: optical-excitatory group (CHR2 group, n=5), optical-inhibitory group (eNpHR group, n=5), optical-control group (O-CON group, n=5); chemogenetic-excitatory group (hm3Dq group, n=10), chemogenetic-inhibitory group (hm4Di group, n=10) and chemogenetic-control group (C-CON group, n=10). The optogenetic or chemogenetic techniques were used in each group. Three weeks after injecting the rat virus, anesthesia was induced and maintained with 2.7% sevoflurane anesthesia in 1.5 L/min O 2, and the EEG data were continuously recorded throughout the process. The burst suppression ratio (%BSR) was recorded at 2 min before and of laser stimulation. Combining optogenetic and chemogenetic strategies, it was investigated that whether activation of orexinergic projection to DRN could modulate anesthetic behaviors during sevoflurane anesthesia. Results:Compared with C-CON group, the recovery of righting reflex (RORR) time was significantly shortened after sevoflurane anesthesia in hm3Dq group ( P<0.05), and the RORR time was significantly prolonged after sevoflurane anesthesia in hm4Di group and eNpHR group ( P<0.05). Compared with O-CON group or the baseline at 2 min before light stimulation, the %BSR was significantly decreased during 473nm laser stimulation in CHR2 group ( P<0.05), and no statistically significant change was found in the %BSR during 473nm laser stimulation in eNpHR group ( P>0.05). Compared with O-CON group, the RORR time was significantly shortened after sevoflurane anesthesia in CHR2 group ( P<0.05). Conclusions:Lateral hypothalamic area orexin-DRN neural circuit plays a key role in promoting arousal from general anesthesia in rats.

ABSTRACT

Objective:To investigate the role of stress-inducible protein Sestrin2 (Sesn2) in the improvement of insulin resistance in rat L6 skeletal muscle cells treated with liraglutide. Methods:The establishment of insulin resistance model of rat L6 skeletal muscle cells was induced by palmitate. The experimental cells were divided into control group(Con group), palmitate 0.6 mmol/L treatment group(PA group), palmitate 0.6 mmol/L+ liraglutide 10 nmol/L treatment group(PA+ Lir10 group), palmitate 0.6 mmol/L+ liraglutide 100 nmol/L treatment group(PA+ Lir100 group), and palmitate 0.6 mmol/L+ liraglutide 1 000 nmol/L treatment group(PA+ Lir1000 group). The cell counting kit 8(CCK8) method was used to detect the cell activity in each group. Western blotting was used to detect the expression levels of glucose transporter 4(GLUT4), protein kinase B(Akt), phosphorylated protein kinase B(p-Akt), and Sesn2 protein in L6 cells. L6 cells were transfected with siRNA to inhibit the expression of Sesn2. The cells were treated with palmitate and liraglutide. Western blotting was used to detect the expression levels of Sesn2, Akt, p-Akt, and GLUT4 protein in L6 cells. Results:Compared with Con group, the cell survival rate, p-Akt/Akt ratio, Sesn2, and GLUT4 protein expression in PA group decreased significantly( P<0.05). After liraglutide intervention, the cell activity, p-Akt/Akt ratio, Sesn2, and GLUT4 protein expression of PA+ Lir100 and PA+ Lir1000 groups was increased( P<0.05). After inhibiting the expression of Sesn2, p-Akt/Akt ratio and GLUT4 protein in transfected si-Sesn2 and treated with 0.6 mmol/L palmitate group(PA+ si-Sesn2 group) and transfected si-Sesn2 and treated with 0.6 mmol/L palmitate+ liraglutide 100 nmol/L group (Lir100+ PA+ si-Sesn2 group) were significantly lower than those in transfection negative group (si-Con group; P<0.05). Even after liraglutide intervention, compared with PA+ si-Sesn2 group, p-Akt/Akt ratio and GLUT4 protein expression level were not significantly increased in Lir100+ PA+ si-Sesn2 group ( P>0.05). Conclusions:Palmitate could induce the decrease of p-Akt/Akt ratio and GLUT4 protein expression in L6 cells. Liraglutide upregulates the expression of Sesn2, which leads to the increase of p-Akt/Akt ratio and GLUT4 protein expression and contributes to the improvement of insulin resistance.

ABSTRACT

The development of immune checkpoint inhibitors (ICIs) has significantly improved the prognosis of cancer patients, but a large population of patients are still ineffective to ICIs treatment or develop aquired drug resistance. In order to improve the clinical benefits, a number of studies on ICIs based combination therapy have been actively explored, and have achieved satisfactory results. With the application of ICIs based combination therapy in clinical practice, increasing attention has been paid to the safety of combination therapy and the management of treatment-related adverse events. In this review, the characteristics of adverse events related to ICIs based combination therapies, especially programmed cell death protein 1/protein ligand 1 (PD-1/PD-L1) inhibitors are discussed, in order to provide profound thoughts for toxicity evaluation and individualized treatment decision in future clinical practice.

ABSTRACT

Objective@#To investigate the relationship between the magnesium intake and patterns of diary and the risk of type 2 diabetes in Harbin residents.@*Methods@#On April 2010, 24 communities in 7 districts of Harbin were selected as research sites using multi-stage stratified random cluster sampling method. A total of 9 734 residents aged 20-74 years was investigated using general questionnaire survey, dietary survey and biochemical indicators test. A total of 9 376 subjects were included in the study. Factor analysis was used to analyze dietary patterns. According to the quartile of dietary magnesium intake, the subjects were divided into four groups, from Q1 to Q4 group. The multivariate logistic regression model was used to analyze the relationship between dietary magnesium intake and the risk of type 2 diabetes within different dietary patterns.@*Results@#A total of 998 subjects with type 2 diabetes were screened. The median age of the diabetic group and the non-diabetic group were 54.8 and 50.8, respectively, and the males accounted for 43.4% (2 896 cases) and 34.6% (433 cases), respectively. The magnesium intake median (P25, P75) of two groups was 336.36 (257.31, 440.65) and 339.50 (264.51, 443.78) mg/d. Four dietary patterns were identified as western dietary mode, savvy dietary mode, traditional dietary mode, and staple food mode. In the western dietary model, the Q4 group had a higher risk of type 2 diabetes than Q1 group, with an OR (95%CI) value of 1.56 (1.06 to 2.32). However, in the savvy diet mode, compared with the Q1 group, the risk of diabetes in the Q4 group was lower, and the OR (95%CI) value was 0.61 (0.37 to 0.96). There was no statistically significant association between dietary magnesium intake and the risk of type 2 diabetes without considering dietary patterns (P>0.05).@*Conclusion@#Dietary magnesium intake has a different relationship with the risk of type 2 diabetes within different dietary patterns.

ABSTRACT

Pancreatic cancer is a highly malignant tumor of the digestive system, also known as "the king of cancer" . Its incidence is increasing year by year worldwide. At present, there is still a lack of effective screening methods for pancreatic cancer, and the early symptoms are not obvious. Most pancreatic cancer is diagnosed in the late stage, and the best time for surgery has been lost, and patients often have poor response to radiotherapy, chemotherapy and targeted therapy, so the prognosis is very poor. The occurrence and development of pancreatic cancer are closely related to genetic background, environmental factors, basic diseases and living habits. So far, although certain risk factors have been identified, such as smoking, obesity, alcohol, chronic pancreatitis, type 2 diabetes mellitus, and family history, the cause of pancreatic cancer is still not very clear. In recent years, more and more studies have shown that in addition to the already recognized risk factors for pancreatic cancer, there is a certain relationship between digestive microecological disorders and the progression of pancreatic cancer. The authors review the research status of digestive system microecology and pancreatic cancer, in order to understand the role of digestive system microecology disorders in the occurrence and development of pancreatic cancer, thus providing a new way to effectively improve the prognosis of pancreatic cancer.

ABSTRACT

Two strains of porcine reproductive and respiratory syndrome virus (PRRSV) were isolated in 2006 and 2016 and designated as FZ06A and FZ16A, respectively. Inoculation experiments showed that FZ06A caused 100% morbidity and 60% mortality, while FZ16A caused 100% morbidity without death. By using genomic sequence and phylogenetic analyses, close relationships between a Chinese highly pathogenic PRRSV strain and the FZ06A and FZ16A strains were observed. Based on the achieved results, multiple genomic variations in Nsp2, a unique N-glycosylation site (N³³→K³³), and a K151 amino acid (AA) substitution for virulence in the GP5 of FZ16A were detected; except the 30 AA deletion in the Nsp2-coding region. Inoculation experiments were conducted and weaker virulence of FZ16A than FZ06A was observed. Based on our results, a 30 AA deletion in the Nsp2-coding region is an unreliable genomic indicator of a high virulence PRRSV strain. The Nsp2 and GP5 differences, in addition to the virulence difference between these two highly pathogenic PRRSV strains, have the potential to be used to establish a basis for further study of PRRSV virulence determinants and to provide data useful in the development of vaccines against this economically devastating disease.

ABSTRACT

Objective To understand the personality characteristics of excellent and generalnurses, extract personality characteristics from excellent nurses, and to establish Personality Characteristics Questionnaire and post personality model of nurses, and explore influencing factors of nurses personality characteristicsfor providing a theoretical reference in selection and training of nurses. Methods A total of 523 staff nurses from one class 3-A hospital general hospitalsin Shandong province were investigated with Sixteen Personality Factor Questionnaire(16PF). Results Excellent nurses possessed higher dominance, rule-consciousness, social boldness, shrewdness, perfectionism and lower apprehension than general nurses (t=-2.54-2.92, P<0.05), then six personality characteristics were extracted from excellent nurses to establish Personality Characteristics Questionnaire and post personality model of nurses; The difference was statistically significant in perfectionism with different age, work experience, and professional title in excellent of nurses (F=4.04-7.36, P < 0.05). Conclusions 16PF is suitable for evaluating personality characteristics of nurses, the personality characteristics questionnaire and post personality model can be used as selection criteria for new nurses.

ABSTRACT

Objective To investigate the difference of the expression of hypoxia reaction genes of vascular endothelial growth factor(VEGF),erythropoietin (EPO) and hypoxia-inducible factor-1 alpha (HIF-1α)) at different times of rats that were induced epilepsy by kainic acid (KA),and analyze their correlation. Methods The epileptic rat model was established by intraperitoneal injection of kanic acid. The expression of VEGF,EPO and HIF-1α gene were detected by real-time fluorescent quantitative PCR assay with TaqMan probe in different times after intraperitoneal injection of KA.Results Compared with normal sodium (NS) group, the expression VEGF was higher at 12 h((8.38±1.27) ×10-3 ng/μl,P<0.05)) and 24 h((8.30±5.08) ×10-3 ng/μl,P<0.05)),EPO was higher at 12 h((8.42±0.90) × 10-5 ng/μl,P<0.05)) and 48 h ((1.50±3.25) × 10-2 ng/μl,P<0.01)) while the HIF-1α was higher at 24 h((2.11±0.21) × 10-2 ng/μl,P<0.01)),48 h((1.50±0.33) × 10-2 ng/μl,P<0.05))and 72 h((1.64±0.16) × 10-2 ng/μl,P<0.01)).Furthermore,the expression of EPO showed significant correlations with HIF-1α and VEGF (r=0.573,0.471,P<0.05),VEGF and HIF-1α had eminent correlations (r=0.803,P<0.01).Conclusions The expression of VEGF,EPO and HIF-1α participate in the seizure procedure and there is certain correlation between the three genes.
